Shuluo,a compound of Chinese medicine,was an original creative drug extracted from herbalist doctors'experience in our lab. Previous experiments has proved Shuluo has the protective effect on cerebral ischemia. In our lab,we had found that coronary blood flow of isolated rabbit heart can be significantly increased by Shuluo.We used acute myocardial ischemia models,myocardial ischemia and reperfusion model , hypoxia/reoxygenation model and cell apoptosis model of HUVECs to study the antimyocardial ischemia/reperfusion effects of Shuluo.In the first part of this thesis, the acute myocardial ischemia models of Pit induced and acute blood stagnation model in rats and MIRI model were established to carry on our research. It was found that Shuluo could reduce the changes of T waves in rat cardiogram. Shuluo could control the platelet and erythrocyte conglomeration, prolong APTT, TT and PT, enhance erythrocyte deformability,and reduce the content of FIB, blood and plasma viscosity. At the same time, Shuluo had the adjustive effect on endothelial system by reducing ET and increasing NO, decreasing the release of CK and LDH. In addition,the pathologic changes including cardiac dropsy,engorgement and soakage of inflammatory cells could be lightened by Shuluo. So Shuluo had protective effects on myocardial ischemia/reperfusion injury mainly by reducing the release of CK and LDH,adjusting the balance of ET and NO and improving the pathological changes of cardiac tissue and hemorheology.The second part was to investigate the mechanisms of Shuluo on myocardial ischemia/reperfusion injury. The HUVECs hypoxia/reoxygenation model was established to investigate the protective effects of Shuluo on HUVECs injury,based on the criteria of the morphologic character, the survival ratio, the intracellular LDH release and the level of ET,NO and NOS.
