Objective To investigate the expression of nuclear factor-kappa B(NF-κB)in hepatic ischemia-reperfusion injury of SD rats and the protective effect of anisodamine. Methods 130 healthy SD rats were randomly divided into shame operation group (group A), ischemia-reperfusion group(group B), normal saline group (group C) and anisodamine group(group D). The SD rats model of hepatic ischemia-reperfusion injury was developed according to Pringle. The expression of NF-κB) in liver tissue was determined by immunihistochemical staining, and Alanine Aminotransferase(ALT), Maleic Dialdehyde(MDA), Superoxide Dismutase (SOD) levels were measured at 45min, 1h, 3h and 6h after reperfusion, and liver tissue samples were observed, with light microscope at 45min, 1h, 3h and 6h after reperfusion. Results The expression of NF-κB and the levels of ALT, MDA increased significantly (P<0.01), SOD decreased significantly (P<0. 01),and necrosis of hepatocytes was visualized in hepatic ischemia-reperfusion injury. There was no marked expression in shame operation group. After treatment with anisodamine, the abnormal changes of all above parameters were diminished remarkably(P<0. 01). Conclusion NF-κB) maybe involved in hepatic ischemia-reperfusion injury. Anisodamine has preventive effects against hepatic ischemia-reperfusion injury by inhibiting the activation and expression of NF- κ B, reducing the production of the free radical, enhancing the ability of eliminating free radical .
