Experimental Study Of Influence Of Thalidomide On Cancer Cachexia Mice

Objective:To establish an experimental cancer cachectic model and observe nutritional state (body weight; left gastrochemius weight and serum concentration of glucose;total protein;cholesterol) and cytokine disorder (TNF-α and IL-6).To observe the effect of thalidomide on cancer cachexia and research the mechanism in cytokine level(TNF-α and IL-6).Methods:1,20 male BALB/c mice were randomized into two groups:no tumor group(NTB) and tumor group(TB). In tumor group, a suspension of 10~6 murine H22 liver carcinoma cells was inoculated s. c into each mouse. Life performance;food consumption and body weight were measured daily after inoculation. Tumor weight;left gastrochemius weight;concentration of glucose;total protein;cholesterol;TNF-α and IL-6 in serum were determined after the mice of tumor group had cancer cachexia.2,70 male BALB/c mice were randomized into six groups: no tumor group (NTB) ; tumor with normal saline group(NST) ; tumor with medroxyprogesterone group(MPA); tumor with low-dose thalidomide group (LTLD);tumor with middle-dose thalidomide group (MTLD) and tumor with high-dose thalidomide group(HTLD). Except the NTB group, the other groups were established experimental cancer cachectic models, then intraperitoneal infusion was given once every day for ten days. The NST group was given normal saline;The MPA group was given medroxyprogesterone solution;The LTLD group was given low-dose thalidomide(50mg/kg) suspension;The MTLD group was given middle-dose thalidomide(200mg/kg) suspension;The HTLD group was given high-dose thalidomide(400mg/kg) suspension. When the treatment was over, some mice were executed and others were observed the survival time.
