In order to increase the bioavailablity of peptides drug preparation for oral administration, recombinant analgesic peptide (rAGP) was chosen as model drug. Pellets of rAGP were prepared for oral administration.It's expensive, fussy and low yield to extract analgesic peptide (AGP) from the Chinese scorpion Buthus martensii Karsch. For the purpose to enhance the yield of AGP, rAGP was cloned by the rAGP-pNJUTRX expression vectors in the E. coli BL21 (DE3). Then expression was induced by IPTG. The recombinant AGP was in soluble form and the expression yield was 12%. The analgesic effect of expressed rAGP was same to extracted AGP. Expressed rAGP was purificated by Chelating Sepharose 4B Fast Flow, then the purity of rAGP was 70%. It was ready to prepare oral formulation.The solubility of rAGP in water was determined by BCA method. The result showed the solubility of rAGP was 0.09mg/ml. In order to improve it, some kinds of solubilizers were chosen. PEG-400 and Tween-80 were better solubilizers, and the solubility of solubilized rAGP was up to 0.94 mg/ml.Multiple preparation means were employed combinatively. rAGP was...
