DC Function Abnormity In HBV Infection And Curative Effect Of HBsAg On It

Abjective Separate dentritic cells from the peripheral blood of patients infected with CHB. Culture and induce DC with AIM-V added with GM-CSF,IL-4,IFN-α. Induce DC with HBsAg inorder to make them producing cyto-immunoreaction target HBV infaction. Measure DC function with ELISA,MTT,immunochemistry,co-culture DC,same person's Tlymphocyte and 2.2.15 cell,observe secretion and growth depression of 2.2.15 cell after Recombinant HBsAg stimulation. Discuss the influence of HBsAg on DC and curative effect of HBsAg in HBV infaction. Seek new approach of chronic HBV infaction treatment.Method Collect peripheral blood of 30 CHB patients and 20 health people and Separate PBMC from peripheral blood with Ficol. Distribute them into three groups:Reccombinant HBsAg treat group(group A),patients group(group B)and the control group(group C). Culture and separate DC with AIM-V(added IFN-α,GM-CSF,IL-4) invitro.
