Objective:To study in vivo gene therapeutic effects of recombinant plasmid pcDNA3.1/hFasL to Graves'disease mice established by immunning syngeneic BALB/c mice with human thyrotropin receptor-activated splenocytes.Methods:Phase 1: 50 BALB/c mice were randomly divided into two groups:Group A of 40 mice immunized with plasmid pcDNA3.1/hTSHR and Group B of 10 mice immunized with plasmid pcDNA3.1. All mice were immunized repeatly after 3 weeks and 6 weeks thereafter. During the period of breeding, two mice from Group A died. Other 48 BALB/c mice were immunized with splenocytes extracted from Group A mice and Group B mice respectively at 18 week.
