Asthma is a global disease which disturbs researchers and seriously effects patients'life quality. However, the incidence of asthma still rises in recent years. Glucocorticoid is widely used for asthma, but problems follow as glucocorticoid-resistance and drug side-effects. Plenty of researches show that the imbalance of Th1/Th2 may be an important factor to induce asthma, while Th2 cytokine IL-13 and Th1 cytokine IL-4 take great part in the progress of asthma. Some researches have showed that neutrolization antibodies and receptor antagonists could blockage their biologic activity. Therefore, IL-13 neutrolization antibody and IL-4 receptor antagonist may play a conspicuous role in the prevention and treatment of asthma in the future.Objective:⑴To establish an asthmatic model of mice.⑵To investigate the effects of IL-13 neutralization antibody on the airway inflammation and Th1/Th2 cell function after acute attack of asthmatic mice.⑶To clone mouse interleukin 4 (mIL-4) truncated gene, construct its eukaryotic expression plasmid pFB-mIL4 and express the truncated protein (murine IL-4 receptor antagonist).Methods:⑴16 BALB/c mice were randomly divided into normal control group and asthmatic model group. Mice in the asthmatic group were sensitized with ovalbumin (OVA) on day 1 and 14, and challenged with 5% OVA on day 21 for 14 days to establish asthmatic model, while mice in the control group were treated with saline instead of OVA.⑵24...
