OBJECTIVES: To study the effect of Skp2 and P27kip1 in development of colorectal cancer through detection the expression of Skp2 and P27kip1 in colorectal cancer and the change of expression of Skp2 and P27kip1 treated by Skp2 antisense oligodeoxynucleotides (ASODN). To investigate the feasibility of genetic therapy of targeting Skp2 for colorectal cancer through the inhibited effect of proliferation of colorectal cancer cell line SW480 treated by Skp2 ASODN.METHODS: Skp2 and P27kip1 protein were examined by using immunochistochemical staining method in 20 cases of normal colorectal mucosa, 20 cases of colorectal adenomatous polyp and 60 cases of colorectal cancer respectively, and analysed the relationship with clinicopathologic features. The liposome-mediated synthetic ASODN encoding Skp2 and nonsense oligodeoxynucleotides(NSODN) were delivered to the colorectal cancer cell line SW480, then the inhibited effect of proliferation of colorectal cancer cell were measured by methylthiazolyl tetrazolium (MTT) methods.The cell cycle were detected by flow...
