Aim: To detect osteoclast differentitation factor levels in synovium tissues from adjuvant-induced arthritis and investigate the role of it in bone and joint destruction during the development of adjuvant-induced arthritis in mature rats.To investigate the protective effect of osteprotegerin on the joints of adjuvant-induced arthritic rats and its possible mechanism.Methods: Sprague-Dawley mature rats were injected with complete freund's adjuvant to induce adjuvant-induced arthritis. On each of days 0,3,7,14,21,30 and 40 after injietion, six rats were killed and the synovium tissue was dissected from knee joints space immediately.Semi-quantitative RT-PCR and Immunohistochemistry were used to detect the expression of osteoclast differentitation factor (ODF) in the synovium.At the same time,Semi-quantitative RT-PCR was used to detect the expression of the osteoclast markers,tartrate-resistant acid phosphatase(TRAP) and cathepsinK, TRAP staining was used to detect the number of osteclasts and hematoxylin and eosin staining was used to evaluate cartilage damage and bone erosion.After two weeks of immunization with complete freund's adjuvant,the rats were randomly divided into two groups.One group was injected osteprotegerin(OPG) into joint cavities one time of each week for 3 weeks,the other was injected with IgG1.After 3 weeks,Semi-quantitative RT-PCR was used to detect the expression of TRAP and cathepsinK in the synovium tissue,TRAP staining was used to detect the...
