Objectives 2, 5-hexanedione(HD),as an active metabolite of industrial organic solvent n-hexane, is believed to mediate the neurogeneration which ensures as a result of n-hexane exposure. The exact mechanism of HD-induced neurodegeneration remains far from elucidated. In this study, we examined the time-dependent alterations of peripheral nerve electrophysiological parameters, oxidative antioxidative system in nerve tissues and serum of HD subchronically intoxicated rats, in order to determine the role of lipid peroxidation in this disease.Methods1. 100 adult male wistar rats were divided randomly into one control and four experimental groups, each 10 animals. HD was administered by intraperitoneal injection to experimental groups with a daily dose of 400 mg/kg.bw for continuous 8 weeks( 4 times per week).2. Body weights and neurobehavioral index were determined per week. Observations of spontaneous locomotion in an open field were evaluated by gait score.3. Determination of nerve conduction velocity, compound action potential amplitude, and other electrophysiological parameters were conducted by in vivo and in vitro testing of anesthetized rats in sciatic nerves at five endpoints (i.e 0,2,4,6,8weeks after intoxication).4.
