Objective: To explore the development and function after transplantation of the fresh and frozen-thawed and cultured human fetal ovary in nude-mice; To investigate the feasibility for hetero-transplantation of human fetal ovary. These findings offer necessary experimental data for organic replacement therapy of some gynecological diseases such as premature ovarian failure.Methods: Human fetal ovary of 20 to 28 weeks were grafted into subcutaneous of female nude-mice for 6 weeks in 6 groups which the fetal ovaries have different treatment such as cryopreserved or cultured. The nude-mice have ovariectomy before transplantation. The recipients" vaginal epithelial cell change was mornitered every day during transplantion. The serum E2 of nude-mice was measured by radioimmunoassay (RIA) and morphorlogic change of fetal ovaries and endometrium of recipients were observed.Results: 1. Vaginal epithelial cell change : normal periodic change was observed in O group; large quantity of premature epithelia in group A ,which means estrogen deficiency; more than 50% mature epithelia was observed in group B,C,D and E group. 2. Serum E2 : The serum E2 of nude-mice in B,C,D and E group was significantly greater compared with A group (P<0.001); there was no significant difference of serum E2 level between the fresh and frozen-thawed group(P>0.05) ; for frozen-thawed ovary tissue the E2 level of cultured group was significantly higher than non-cultured group(P<0.001). 3.
