Objective: Lung cancer is one of the most common malignant tumors in human, and the arising, development of lung cancer has strong relationship with smoking. It is important to investigate the mechanisms of lung carcinogenesis for developing the diagnostic and therapeutic approaches. Tob is one member of the Tob/BTG family, a novel class of anti-proliferative gene. We analyzed the expression of tob mRNA , the expression of Tob protein and the phosphorylated form of Tob protein(an inactive form of Tob) in lung cancer tissue and adjacent normal lung tissue, and studied their relationships with smoking. To investigate the involvement of tob as a tumor suppressor gene in human lung cancer, to get the relationship of characteristic of clinical pathology and smoking.To understand the mechanism of lung cancer arising , further we can provide theoretical basis of cancer control, prevention and genetic diagnosis and treatment in early stage of lung cancer.Methods: 36 lung cancer patients were randomly selected, samples of tumor and normal lung tissues were collected during operation, then stored freshly in -80°C freezer and fixed in 10% formalin separately. For each patient, clinical history was detailed collected including smoking history before operation, and pathological characteristics after operation, then the following methods were used to study the expression or phosphrylation status of tob in lung cancer and their relationship with smoking: (1) Total RNA was isolated from tissue by RT-PCR, the level of tob mRNA was determined by tob/β -actin ratio.(2) Immunohistochemiscal SABC technique was applied to study the expression of tob protein or...
