Objective To observe the pathlogical changes and investigate the hepatocyte apoptosis relating with CYP2E1 and oxygen free radical in alcoholic liver disease(ALD) of rat.Method The dose of 40% ethanol was administerd at 8g/kg body weight by gavage twice daily for 8 weeks in model group, and control group was recived isovolume saline by gavage. End of 8th week, the the activities of serum ALT and AST were detect by autobiochemistry instrument. The pathological changes of liver was observed under light microscope after HE staining, and hepatocyte apoptosis was detected by the TUNEL method. Exprresion of serum CYP2E1 was determined by polymerase chain reaction(PCR) method, the contents of Maleic Dialdehyde (MDA) and activities of SOD in serum were detected by Thibabituric Acid (TBA) quantifyingmethod.Results (1)Compared with that in the control group, the level of serum ALT and ASTwas significantly increased (107.4±16.6 vs 48.4± 10.5IU/L, 232.1 ± 52.9 vs 102.9 ± 14.1IU/L, (P<0.05), the content of MDA and activities of SOD in serum were significantly increased (1236.4 ± 283.5 vs 745.6 ±174.8ng/L; 41.53 ± 7.43 vs 15.72 ± 2.06nmol/ml,636.82 ±138.60 vs 353.12 ± 61.02 u/ml, all P<0.05) in the ALD group.(2)Compared with that in the control group, the hepatocytes became swelling and macro- and microvesicular stratosis , spotty and focal necrosis,and inflammation were observed, the hepatocyte apoptosis was significantly incresed (P<0.05), and the TUNEL positive cells were located around the central vein, spotty and focal necrosis in the ALD group..
