Objective To investigate the proliferation and differentiation of bone marrow-derived mesenchymal stem cells(MSCs) when transplanted into post-infarct myocardium and their influences on cardiac function. To compare the effects of the three different ways of autologous bone mesenchymal stem cells manipulation in vitro on post-infarct myocardium, selecting the optimal way of stem cells treatment in vitro.Methods .Myocardial infarct model was set up by ligation of the left anterior descending coronary artery ( LAD ), Two weeks after the ligation, fourty New Zealand rabbits myocardial infarct models were randomly divided into four groups: cultured bone marrow-derived mesenchymal stem cells (MSCs, n=10), 5-aza-treated bone marrow-derived mesenchymal stem cells (5-aza-treated MSCs, n=10) , fresh bone marrow mononuclear cells (BMMNCs, n=10) which labled with 1,1'-dioctadecyl-3,3,3',3'-tetramethyl indocarbocyanine perchlorate (DiI), and medium (control, n=10) ,were autologously transplanted into several different points of the border of the scar respectively.To evaluate the heart function , echocardiography were performed before the LAD ligation ,two weeks after the ligation, two and four weeks after the cells transplantation for (1) M type for measurements of left ventricular end systolic diameter (LVESD) ) and left ventricular end diastolic diameter (LVEDD),to calculate left ventricular eject fraction(LVEF)and fractional shortening (FS),(2) Doppler Imaging to obtain the blood flow velocities at the orifices of aortic valve,pulmonary valve, mitral valve,and tricuspid valve; Doppler Tissue Imaging(DTI) were performed for measurement of the velocities of the ring of the mitral valve, Meanwhile the myocardial contrast echocardiography (MCE) were performed for evaluating the blood perfusion of the post-infarct myocardium.
