The Effects About The Cyclic Pressure On Constructing Of Tissue Engineering Cartilage In Vitro

Objective:We explore the effect of the cyclic pressure on constructing of tissue engineering cartilage in vitro and carry on an initial research about its mechanisms. Methods:Kill the New Zealand rabbit, sterilize the skin with Iodophors. Articular cartilage was taken from the rabbit under sterile environment. And shear it to the size of about 1mm×1mm×1mm. Then chondrocytes were isolated from the articular cartilage by digesting with 0.25% trypsin and 0.2% type II collagenase in sequence. After expansion in vitro, the 4^th passage chondrocytes were seeded into PGA scaffolds and cover slips. After adherence, the scaffolds and cover slips were both divided into 2 groups. Experimental group was subjected to the cyclic pressure, while the control group was cultured under normal condition. Both groups were cultured in vitro. After 5 days, the concentration of calcium within chondrocytes was detected with the use of the laser confocal microscopy.
