Objective:We explore the effect of the cyclic pressure on constructing of tissue engineering cartilage in vitro and carry on an initial research about its mechanisms. Methods:Kill the New Zealand rabbit, sterilize the skin with Iodophors. Articular cartilage was taken from the rabbit under sterile environment. And shear it to the size of about 1mm×1mm×1mm. Then chondrocytes were isolated from the articular cartilage by digesting with 0.25% trypsin and 0.2% type II collagenase in sequence. After expansion in vitro, the 4th passage chondrocytes were seeded into PGA scaffolds and cover slips. After adherence, the scaffolds and cover slips were both divided into 2 groups. Experimental group was subjected to the cyclic pressure, while the control group was cultured under normal condition. Both groups were cultured in vitro. After 5 days, the concentration of calcium within chondrocytes was detected with the use of the laser confocal microscopy.
