Objective To explore the effects of proteasome inhibitor PS-341, As2O3,Dexamethasone(Dex) and Thalidomide(Thal) on the abnormal expressions of cytokines of mesenchymal stem cells(MSCs) in patients with multiple myeloma (MM) .Methods Bone marrow MNCs of 11 patients with multiple myeloma were separated by Ficoll and cultured in medium of 1640 containing 10% FBS. After growth for 2-4 generations , MSCs were determined by inverted light microscopy and flow cytometry analysis, then MSCs were exposured to PS-341, As2O3, Dex or Thal for 4h, 8h, 8h, 8h, respectively and detected the expressions of IL-6,IL-1β, SCF by reverse transcription PCR(RT-PCR). The results were analysised using t test. Results The expression levels of IL-6, IL-1β and SCF decreased markedly after treatment with 50nmol/L, 100nmol/L, 150nmol/L, 200nmol/L PS-341, 0.2μmol/L, 0.4μmol/L, 0.6 μmol/L, 0.8μmol/L As2O3, 0.1μmol/L, 1.0μmol/L, 5.0μmol/L, 10.0μmol/L Dex and 1.0μmol/L, 3.0μmol/L, 5.0μmol/L, 7.0μmol/L Thali for 4h,8h,8h, 8h, respectively ( P<0.05 ) .PS-341 inhibited the expressions of IL-6, IL-1β and SCF of...
