The Experimental Research Of Adenovirus-Mediatedtransfer Of IL-12 Gene On The Mouse Surrounded By Liposome Transplanted Kupffer Cells

Objective:(1) To isolate and culture KCs from SD mouse liver and receive pure and active KCs which can be used for further experiment. (2) To construct recombinant adenovirus -mediatedtransfer of IL-12 gene on the mouse(Adv-mIL-12) which can express IL-12 highly and stably and make it possible for gene therapy. (3)To detect the expression of IL-12 by KCs which have included Adv-mIL-12 surrounded by liposome through phagocytosis action and to provide the evidence of targeted therapy on micrometastasis liver cancer in such way.Methods:(1) nonparenchymal cells can be isolated from parenchymal cells in the mouse liver through ex vivo perfusion using typeâ…£collagenase and tautologically centrifuge.
