ObjectiveTo observe the prevention and treatment effects of celccoxib on implanted human primary hepatic carcinoma of nude mouse, and study the mechanisms of the effects. Materials and MethodsForty nude mice, 4 weeks old, were used. The cell, line of human primary hepatic carcinoma, namely SMMC-7721, was cultured at 37℃ in DMEM medium, supplemented with 15% calf serum. Tumors were established by injection of 4×10~6 tumor cells into the back of nude mice, and growth determined by measuring the long and short diameters.The mice were classified randomly into four groups consisting of 10 mice each, one group as control, two groups receive the diet supplemented with celecoxib of two different concentration starting 3 days prior to tumor cell injection and throughout the whole experiment. One group receives the diet supplemented with a high concentration celecoxib, starting when the tumors reached 0.2cm in diameter. To observe the tumor growth curve, and obtain the tumor samples. The tumor samples were used in the examinations of the expression of VEGF, caspase3, caspase9, MMP2 with immunohistochemistry and MVD in tumor with immunohistochemistry, and of cell-apoptosis with TUNEL staining.
