[Objective]①The domestic recombinant human AnnexinV is labeled with *I by using Iodogen method. To investigate the in vivo distribution and detect the pharmacokinetic parameters of *I-AnnexinV with the mice bearing S180 sarcoma tumor. To explore the feasibility of imaging apoptosis in tumor with a new apoptosis imaging agent radioiodin labeled AnnexinV(*I-AnnexinV).②To investigate the clinic value of *I-AnnexinV and 99Tcm-MIBI dual-isotope imaging for assessing tumor apoptosis in mice bearing S180 Tumor.③To estimate the human radiation absorbed doses of 123I-AnnexinV, 125I is used as a radiotracer for measuring the distribution of radiolabeled AnnexinV in mice.[Methods]①AnnexinV is labeled with 125I.②125I-AnnexinV with the dose of 37MBq/kg are injected into 45 Kunming mice bearing S180 sarcoma via tail vein. The animals are sacrificed at 0min, 5min, 10min, 30min, 1h, 3h, 6h, 10h, and 24h after injection. The heart, lung, kidney and other organs are obtained, weighed and counted in a gamma scintillation counter for 125I activities. The in vivo distribution of 125I-AnnexinV in mice and its pharmacokinetic parameters are calculated with DAS 2.0 soft ware.③One group of mice bearing S180 sarcoma tumor is treated with cyclophosphamide to induce apoptosis of sarcoma. The other group is injected with 0.9% normal saline.④Kunming mice bearing S180 sarcoma is used for SPECT imaging and ROI technique is adopted to measure the T/M. At the same time, *I-AnnexinV and 99Tcm-MIBI dual-isotope imaging are used to evaluate apoptosis in mice bearing S180 sarcoma tumor.⑤Tumor biopsies are collected immediately after SPECT imaging.The expression...
