But the improvement of immature oocyte cryopreservation was very slow, immature oocyte cryopreservation so far has been applied with limited success and there is no unified standard about cryopreservation methods.To achieve oocyte cryopreservation,the two approaches of controlled slow-rate freezing and rapid vitrification have been developed . Many experiments showed that slow-rate freezing of mature oocyte could bring damage on the meitotic spindle;but about the immature oocyte,the results were not identical.and Many experiments showed that vitrification of mature oocyte and matured in vitro could bring bring irreversible damage;but about the immature oocyte,the results also were not identical. Which may be the different of species, cryoprotectants and the freezing methods. Now,we havenot known that to the effects of the meitotic spindle,which is the betterfreezing method and the better freezing stage.So It is very important to explore the factors which effect oocyte cryopreservation and find the optimal stage and method.Compared with immunostaining and fluorescenequenching,or other microscopy methods,the Polscope is totally non-invasive,and therefore oocyte viability can be preserved.Repeated oberservations are possible over time.and the Polscope quantifies microtubule architecture better than immunofluorescene,because the latter introduces artifact related to fixation .
