The Effect Of ShenQi Compound Recipe On Apoptosis And Metastasis Of CNE Cells

Objective:The therapies of cancer are still in difficulties in medicine.There are so many ways to treat cancer at present.The study of traditional herbal medicine which are reported to have a good anti-tumor effect is more and profound.ShenQi compound(SQ)which is based on ShenQi tablet is made with addition of Astragalus root?￠chinese angelice root?￠crcuma zdoaria and oldenlandi diffusa.The effect and mechanism of anti-tumor was reported and studies suggested SQ could suppress cancer in vivo and in vitro of mouse.This experiment will do some research on anti-tumor mechanism of SQ further by studies on SQ?ˉs effect on apoptosis and metastasis of cell of human nosapharynx cancer(CNE).It is importance of developing new anti-tumor drug which is more curative and has low side-effect.Methods:CNE cells cultivated in vitro are made to cell suspension,then add SQ of different concentration .24?￠48?￠72 hours later,MTT is used to assay the inhibition rate of CNE cells.After transfer of culture of CNE cells,adjust cell concentration to 5.0?á105/ml.Add SQ of 31.25mg/ml?￠62.50mg/ml?￠125.00mg/ml and NS.48h later,do the experiment following.Extract DNA of tumor cells of experimental group(62.50mg/ml).After 1% agar gel electrophoresis,observe characteristic DNA ladder due to apoptosis.Add the Fas-FITC monocolonal antibody to the SQ treatment group and control group.Flow cytometry is used to determine the level of Fas expression.Extract RNA of tumor cell of 31.25mg/ml and 62.50mg/ml treatment group and control group.Mesure the concentration and degree of...
