| Background The pathogenesis of type 2 diabetes is not clear yet. It is hard to identify type 1 and type 2 diabetes in clinical. Although type 1 and type 2 diabetes belong to two kinds of diseases, there maybe exist a common mechanism inβcell dysfunction. CTLA-4 participates in the control course of T cell immune response. Although CTLA-4 49 closely related to type 1 disease, there is lack of the research about the association between CTLA-4 and type 2diabetes.Objective Search on the association between CTLA-4 gene variation and the type 2 diabetes, compare the meatabolic character of different genotype and explore wether it is a heridity susceptility index to identify diabetes of insulin lack ( type 2 diabetes ) and diabetes of insulin resistance ( type 1 diabetes ).Method Randomly select 111 cases of type 2 diabetes patients (diagnosed as the critera of ADA 1999) and 39 healthy people as control(dispite of typical type 1 diabetes and secondary diabetes). The control group is dispite of the person of IGT, or with diabetes family history or related metabolic diseaseas as well.After routinely fasting 8-12 hours, the participants were attracted blood from periferial venous.The blood were centred to be taken serum and picked up genome DNA from nucleus cell of peripheral blood. The genotype of CTLA-449 was analyzed in patients with type 2 diabetes, and controls were analyzed by polymerase chain reaction-restriction fragment length polymorphism analysis.We detect their blood pressure, FINS, TG, CHOL, HDL-C, body weight, height, LDLC and FBG as well.Calculate BMI with body weight and height. Statistic analysis the data with different groups. |
