Objective: To investigate the correlation of glucocorticoid receptor(GR) gene BclI polymorphism to diabetes mellitus and its metabolic componentsMethod : 194 type 2 diabetic patients were selected according to WHO /1999 criteria. For diabetic patients 2 groups were defined according to IDF 2005 criteria of MS .Group A(DMA) : waist circumference(W)≤90 cm in men and≤80 cm in women, Group B (DMB): waist circumference >90 cm in men and > 80 cm in women.117 subjects with normal fasting glucose(FBG<5.6mmol/L). 2 groups (NFGA&NFGB) were also defined as in diabetic subjects according to IDF 2005 criteria of MS. GR genotypes in intron 2 were determined by polymerase chain reaction restriction fragment length polymorphisms (PCR-RFLP) assay . The frequencies of GR genotype and allele gene were compared. In each group, body mass index(BMI), total cholesterol(TC), triglyceride(TG), high density lipoprotein(HDL-Ch), low density lipoprotein(LDL-Ch),insulin sensitivity index(ISI), systolic(SBP) and diastolic(DBP) were used as metabolic indices for contribution of genotype variation .Results: The overall frequencies of GG, GC, CC in DM and in NFG were 24.2%, 55.3%, 10.5%, 30.6%,60%, and 9.4% respectively. There was no difference in the genotype frequency distribution between diabetes and NFG subjects. The frequencies of GG, GC, CC genotypes in DMA were 30.3%, 60.6%, and 9.1% respectively, while in DMB, 29.8%, 62.7%, and 7.5%. There was no difference between the two groups.
