Metastasis is the major biological feature that benign tumors are different from malignant tumors, it is also an important reason which causes malignant tumor patients to die. Most malignant tumors are cancers which derives from epithelial tissue ,however ninety percent of cancers metastasize by lymph approach. So research in lymphatic metastasis mechanism can help to early diagnosis of tumor and effective therapy. Proteomics has been widespreadly implanted into every field of life science and medicine as an important part of post-geomics era research .It can study lymphatic metastasis as a whole to cell proteome. The two-dimensional gel electrophoresis technology, established by O'Farrel in 1975, has still been the vital techonology in proteomics research depending on its excellent resolving power . Carrier ampholytes two-dimensional gel electrophoresis was used to display and compare the differential expression of lymphatic metastasis related proteins in Hca-F and Hca-P cells and in order to make the foundation for further studying lymphatic metastasis mechanism.Methods The total proteins of Hca-F and Hca-P cells were extracted by ultrasonic wave and chemistry lysis method respectively, and colorimetric method was used to detected their concentration .The total proteins of two cell lines were separated by carrier ampholytes 2-DE and the spots of proteins were visualized by Coomassie brilliant blue. The differentially expressed proteins were analyzed using PDQuest 8.0 software.
