Objective: To investigate the antinociceptive effect of ondansetron on the experimental pain modelMethods:The study was divided into 4 parts .1 The experimental study on antinociceptive effect of ondansetron in local use1) Effect of exposure time on the antinociceptive actions of topical ondansetron in mice: Groups of mice received 0.5, 1.0, 2.0 ,5.0 and 10 min topical exposure to a fixed concentration of ondansetron(32 mg/ml). Tail flick latencies were obtained 30 min following topical exposure.2) Antinociceptive actions of topical ondansetron in mice: Mice were randomly divided into 5 groups ,ondansetron group(2mg/mL,8mg/mL,32mg/mL),Lidocaine group (20mg/mL) and DMSO group(n=8). Groups of mice received a 2 min topical exposure to different concentrations of drugs. Tail-flick latencies for each animal were measured from regions of the tail exposed to the compound or not across a 180 min time period.2 The effect of local Ondansetron injection on pain behavior spinal fos expression in a rat model of incisional painRats were randomly divided into saline, lidocaine, ondansetron and sham operated groups(n=8).Drugs were given locally .15minutes later ,incision was made in the plantar aspect of the left hindpaw .A cumulative pain score was utilized to assess pain behavior within 1h after the incisional pain model was made .The animal were decapitated 2h after incision and the spinal cord was immediately removed and sliced and examined for fos expression using fos immunohistochemistry .3 An electrophysiological study of local ondansetron injection on dorsal horn neuron activityRats were randomly divided into formalin, lidocaine, ondansetron and saline group...
