The prognosis of optic nerve injury is not good, about 40 to 50 percent patient would lose their sight, up to now, there is not any effective therapy for it. At present, it has been proved that in adult mammals, the inhibitors in the surrounding of optic nerve(ON) may be major contributors to the inability of injured neurons to regenerate except for the deficiency of neurotrophic factors, the formation of glial scar, the low regeneration ability of retinal ganglion cells(RGCs). The investigations published in 2002 showed that the three inbibitors in oligodendrocyte and myelin, Nogo-66, MAG and OMgp, produce a marked effect all through the NgR, the monoclonal antibody to NgR could effectively promote the regrowth of central nerve neurite.NgR mRNA is widespread in the central nervous system(CNS), such as neocortex,hippocampal formation, dorsal thalamus, cerebellar granule cells, etc. However, as a special kind of central nerve, wheather or not is there NgR mRNA in the ON? If the answer is yes, can we use RNA interference(RNAi) technology to silence the NgR gene and restrain the function of Nogo-A, MAG, OMgp, then to promote the regeneration of ON?The purposes of the study are: detecting the expression of NgR mRNA in the ON, observing the change of NgR mRNA after the injury of ON, constructing NgR gene specific siRNA encoding vectors, laying the foundation for using RNAi to inhibit the NgR gene and to urge the regeneration of ON.The study was divided into three parts:1.to verify that NgR mRNA is expressing in the ON by using in situ hybridization; 2.to observe the change of NgR mRNA after ON injury by using fluoresence quantive polymerase chain reaction;3.to construct two plasmids which encoding NgR specific siRNA.The main results and conclusions are as follows:1.
