Objective and Significance:The proliferation,differentiation,maturation and apoptosis of the leukemia cells are regulated by the immunological network.The occurrence and development of leukemia are in relation to the dysfunctional immunological regulation and the abnormal change of the hematopoietic inductive microenvironment(HIM).The hepatocyte growth factor(HGF),produced by the bone stromal cells, is an important factor in the hunman immunological network and its abnormal expression may be one demonstration of the HIM. C-met , the HGF's only receptor, is generally expressed in the leukemia cells during pathological situation. Only when the HGF combinds with the C-met ,it can produce a marked effect.In contrast to the normal mononuclear cells(MNCs),the HGF can alone stimulates the proliferation and colony formation of MNCs from leukemia patients, enhance the adhension between the leukemia cellls and BMSCs and induce the migration of the leukemia cells. Those facts indicate that HGF contributes to the occurrence, development and prognosis of leukemia, so it is important to investigate the expression of the HGF in leukemia and its relationship with the occurrence, infiltration and therapeutic efficacy of leukemia. We detected and analysed the expression of HGF in the BMSCs and the serum before and after treatment in order to study the relationship between the HGF and the proliferation and infiltration of the leukemia cells as well as to provide a new helpful experimental index for the treatment and prognosis of leukemia.Methods:43 patients with leukemia includes:16 ANLL patients,9 ALL patients,11 CGL patients,7 CLL patients,15 CR patients;20 patients(rule out malignant tumor and other major diseases) were choosed as controls. Their BMSCs and peripheral blood were collected for detection. The expression of the HGF in the BMSCs was detected by the immunocytochemistry and ELISA; the expression of the HGF in serum was detected by ELISA. The patients'clinical materials considering leukemia classification, infiltration and remission degree were also collected .All the data were analysed by SPSS statistic tool.
