Objective: The purpose of this study was to determine whether potentiating induction of apoptosis in cervical cancer cell line SiHa lacking full-length FHIT transcripts and endogenous FHIT protein by cisplatin via introduction of extopic FHIT gene. Meanwhile, to investigate if the apoptosis of the SiHa cells is induced by FHIT gene.Methods: PRcCMV-FHIT (SiHa-FHIT group) and PRcCMV (SiHa-neo group) were transfected into cervical cancer cell line SiHa Lacking full- length FHIT transcripts and endogenous Fhit protein(blank SiHa cells is called SiHa-C group), the expression of extopic FHIT protein was examinated by Western blot, with different cisplatin, the proliferation inhibition ratio of SiHa cells was examinated by MTT, apoptosis were examinated by AO-EB stain and flour cell assess(FCA).Result: The percentage of transtient transfection were not significantly difference between lipofection and electrofection. The apoptosis rate of SiHa-FHIT,SiHa-neo, SiHa-C group was respectively 22.07%, 4.86%, 3.75%, percentage of G0/G1 phase of SiHa cells transfected by PRcCMV-FHIT was increased (percentage of cells in Go/Gi phase was 63.70%, that of control was 47.24%). The apoptosis rate of SiHa-FHIT, SiHa-neo, SiHa-C group was respectively 51.03%, 20.48%n 20.58%, the proliferation inhibition ratio of SiHa cells was respectively 69.54%, 42.98%, 41.30% when three group cells is treated with 2.0μg/ml cisplatin. The proliferation inhibitory rates and apoptotic rates of SiHa-FHIT + 2.0μg/ml DDP group were significantly higher compared with SiHa-neo+2.0μg/ml DDP group and SiHa-C+2.0μg/ml DDP group (P<0.01) , the difference between SiHa-neo+2.0μg/ml DDP group and SiHa-C+ 2.0μg/ml DDP group wasn't significant (P > 0.05 ).
