AIM:To establish a new method to derive foam cells from macrophages. Making use of it on the study of ezetimibe to find a new mechanism of anti-atherosclerosis.METHODS: The concentration of total cholesterol and free cholesterol were detected by high performance liquid chromatography. Oil Red 0 Dyeing experiment was used to show the cellular lipid droplets in lipid-loaded cells. The expression of SREBP-1 and caveolin-1 were measured by Western blotting.RESULTS:Treatment with 45ug/ml Cho1:MβCD for 72h on RAW264. 7 cells, It was found that the content of cellular cholesterol came to the peak value. There are no significant difference between treatment with 45ug/ml Chol:MβCD for 72h and 50ug/ml ox-LDL for 48h on RAW264. 7 cells. When treatment with 45ug/ml Chol:MβCD, we also added various concentration of eztimibe or 10umol/l lovastatin into medium for 72h. The results showed that ezetimibe decreased the concentration of cellular cholesterol and the amount of lipid droplets in dose-dependent manner. The expression of caveolin-1 and SREBP-1 increased when treatment with Chol:MβCD for 72h. After cotreatment with Chol:MβCD and ezetimibe, the results showed that Chol:MβCD decreased the expression of caveolin-1 and SREBP-1 in dose-dependent manner. Lovastatin also decreased the concentration of cellular cholesterol and the amount of lipid droplets .At the same time , the expression of caveolin-1 and SREBP-1 also decreased. Conclusion:1. Foam cells can be derived from RAW264. 7 cells by Chol:MβCD.2. Ezetimibe can prevent foam cells derived from RAW264. 7 cells by Chol :M-... |
