| Objective: To investigate the effect of curcumin on expressions of p300,c-myc, P53 and actyle-histone H3 and H4 in the SGC-7901 in vitro, and its molecularmechanism of anticancer.Method: After the gastric cell line SGC-7901 was treated with differentconcentrations of curcumin and different cultural time in vitro. The cellular proliferation was evaluated by MTT assay. The mRNA expressions of P300 and c-myc gen were assayed by reverse transcriptive polymerase chain reaction(Rt-PCR).The protein expressions of acetyl-histone H3 , H4 and P300 were tested by western blot. The protein expressions of P53 and C-myc were investigated by immunohistochemical techniques. Results: MTT assay indicated that curcumin could inhibit the proliferation ofSGC-7901 cells line in the time and dose-dependent manner. There was great statistical signification between varies medicated group and control group (P<0.05) , and there also were great statistical signification between medicated group each other (P<0.05) . The mRNA expressions of P300 and c-myc were totally decreased gradually in SGC-7901 cell treated with curcumin in concentration and time dependent mode, there was positive correlation between them. However, the mRNA expression of wide-type P53 was not affected . Curcumin could inhibit the protein expressions of P300, acetyl histone H3 and H4 in concentration and time dependent manner(P<0.05).
