Whooping cough is a serious respiratory disease caused by Bordetella pertussis. However, Pertussis vaccine is the most effective method for pertussis transmission .The side effects of the current whole-cell vaccine are seriously, as a consequence, acellular pertussis vaccine was developed as alternatives to the whole-cell vaccine for its better protective effect and less side effects. Acellular pertussis vaccine recommended by WHO should contain five components: PT, FHA, PRN, FIM2 and FIM3, the major component is PT. The PRN, FIM2 and FIM3 play important roles in pathopoiesis and immunity of pertussis. It has been demonstrated that acellular vaccine containing multiple pertussis antigens (such as PT, FHA, PRN, FIM ) is more efficacious than single-component acellular vaccine.It is not only a hot spot of basic studies of pertussis but also a key point of developing safer and effectiver vaccines in our country that study on the background of the primary seed of producing strain, add more protective antigens in acellular pertussis vaccines and evaluate their protective effects and set up sensitive and specific ELISA methods for the quality control and clinical efficacy evaluation of acellular pertussis vaccines. There are four parts in our study:Firstly, we used pertussis CS strain which are currently used in preparing acellular vaccine in china as the template, amplified gene fragments of PT,FHA, PRN,FIM2 and FIM3 by PCR , and inserted them into PGEM-T easy vector respectively , after identified correctly by Enzyme Digestion and PCR, these gene fragments were sequenced and analyzed. The lengths of these gene fragments were 3139bp, 2740bp, 2272bp, 701bp and 582bp respectively , the open reading frameworks were complete and no sense mutations were found. Homology analysis showed that the homologies of nucleotide and amino acid sequences of the five gene fragements of CS starin were more than 90% compared with those of Bordetella...
