Objectives: Diltiazem Hydrochlorid is abenzothiazepine derivative with calcium-channel activity, which can prevent circadian angina pectoris. According to chronopharmacokine-tics the pellets in this paper were designed to combine with sustained-release pellets and pulsatile controlled-release pellets to prevent and cure heart-blood event.Methods: On the basis of scientific materials and preparation experiment, to preparation sustained-release solid dispersion of DIL with acrylic resin â…¡ to decrease solubility of DIL. The sustained-release pellets were prepared by extrusion-spheronization with microcrystalline cellulose (MCC). The main factors inspected involved factors of prescription (ratio of DIL: â…¡, amount of MCC, dose of adhesive, different size of pellets) and factors of technique (speed of extrusion, speed of spheronization, time of spheroni- zation), which influenced formation and release of pellets.
