ObjectiveTo study the change and signifinance of expand function in the 2 type diabetes mellitus rats′bladder.To investigate the expression ofβ-receptor and G-protein in the 2 type diabetes mellitus rats′detrusor cell.MethodsThe present study is composed of the following three sub-article: Part one :Experimental study of excitability and autorthmicity and contraction/relaxation inurinary bladder of 2 type diabetes mellitus rats(DM).Using theβ-receptor agonist IsoprenalineHydrochloride stimulates the detrusor strip in the different medicine density(1×10-10~3×10-5)and observing the change of relaxation function between the normal and 2 type diabetes mellitus rats.Part two :In the different time digestion detrusor cell applying the Flow Cytometry(FCM)to examine the change ofβ3-receptors.Part three :Making the bladder into homogenate and using the western blot to examine the change of G-protein.Results1.Experiment study of detrusor strip relaxation function :IC50 for DM is more responsive and the Emax is strong in 8 week(P<0.05).However, IC50 and Emax is decrease in 16 ,24week (P<0.01).2.The mean FL-Height ofβ3-receptor is increased of DM group in 8 week(P<0.05). DM group became decreased in 16 week(P<0.01)and it is significant decreased in 24 week(P<0.001).3.Using Westernblot examine the change of G-protein we found that the two group is unvariance for Gsαin 8 week(P>0.05).But the Gsαof DM group became decreased in 16 and 24 week(P<0.05).The Giαis minute and we is difficult to examine it. ConclusionThe relaxation function of DM group rats detrusor is strong which is possible the result of increasing ofβ3-receptors in 8 week.But when the detrusor is decompensation theβ3-receptors and Gsαboth became decreased.In advanced,it may make the signal transmission pathway...
