Objective: Study the protective effects of hypoxic preconditioning on rat myocardial contusion and inquire mechanism.Methods: Rat myocardial contusion model was established ,60 health and mature wistar rats are classified into three groups with no purpose and the rats of every group was partitioned into 2h,4h,6h,12h group. Hypoxic preconditioning model was established through making the rats breath the mixed gas combined with 10%O2 and 90%N2 for 3 hours ,subsequently to the hypoxia the rats was fed for 24 hours under normal condition. The change of myocardial histology was observed by useing HE staining, the expression of cTnT and bcl-2 was detected useing immunohistochemical staining and analysised by computer system of image analysis . Results: (1)There is obviously depletion area of cTnT after 1 hour in myocardial contusion of rat,and the area of depletion increased with the prolongagtion of contusion time. At the same time there is not obvious depletion of cTnT in control group compared myocardial contusion .(2) there is little expression of bcl-2 in control group rat myocardial and the expression of bcl-2 reduced in concardial group, there is obviously enhancement of the expression of bcl-2 in HPC group compared myocardial contusion group and control group.Conclusion: HPC can reduce the degree of myocardial contusion and the protective effect probable inhibit apoptosis of myocardial cell through adjusting the expression of bcl-2.
