Objective:To explore the role of connective tissue growth factor(CTGF) and nuclear factor-κB (NF–κB) in the renal on the process of renal interstitialfibrosis caused by unilateral ureteral obstruction(UUO) and partial unilateral ureter obstruction (PUUO) in rats.Methods:Forty - eight rats were randomly assigned to shame operation group (normal group) ,complete obstruction group after unilateral ureteral obstruction, partial obstruction group after partial unilateral ureter obstruction, sixteen per group. Rats were killed at the 3 rd ,7th ,14th , 21st days after UUO,PUUO and sham-surgery. The protein expression of CTGF and NF–κB in renal was detected by immuohistochemistry at each time point .Results:(1) In comparison with shame operation group , tubular Lesion , interstitial fibrosis ,the expression of CTGF and NF -κB in UUO group markedly increased at 3, 7, 14, 21day.( P < 0.05) . In comparison with shame operation group , tubular Lesion , interstitial fibrosis ,the expression of CTGF and NF -κB in PUUO group markedly increased at 7, 14, 21day.( P < 0.05) .(2) In comparison with UUO group , tubular Lesion , interstitial fibrosis ,the expression of CTGF and NF -κB in PUUO group markedly decreased at the same time point.( P < 0.05) .(3) Tubular Lesion , interstitial fibrosis ,the expression of CTGF and NF -κB in UUO group and PUUO group markedly increased with change of times. ( P < 0.01)(4) The expression of CTGF was significantly positively correlated with tubular Lesion , interstitial fibrosis in UUO and PUUO group. ( P < 0.01) The expression of NF–κB was significantly positively correlated with tubular Lesion , interstitial fibrosis in UUO and PUUO group. ( P < 0.01)(5) The expression of CTGF was significantly positively correlated with that of NF–κB in UUO and PUUO group. ( P < 0.01)Conclusions:CTGF and NF–κB may play a crucial role in the process of renal interstitial fibrosis. CTGF synergistically with NF–κB to induce the development of renal interstitial fibrosis.Clinically,we should indetify the diagnosis and have a operation early.
