OBJECTIVE: To suggest an immune measurement by studying the effect of recombinant rat IL-10 on in vitro IL-2,TNF-αproduction of peripheral blood mononuclear cells of epileptic rats. METHODS:32 rats were randomly divided into four groups. Abdominal cavity injection Bemegride used to produce epilepsy models. Mononuclear cells was separated and purified by lymphoid separating medium.Peripheral blood mononuclear cells of epileptic rats were cultured for 48h and divided into control group and three IL-10 treatment groups.The content TNF-αin supernatant was measured by ELISA. Nested RT-PCR was used to detect the expression of interleukin-2 mRNA.RESULTS: Compared with the control, all the group of IL-10 can inhibit the level of TNF-αproduced by PBMC(P<0.01). But there was no significant difference instatistics between group 20 ng/ml,40 ng/ml(p>0.05). All the group of IL-10 can inhibit the expression of IL-2mRNA produced by PBMC(P<0.01). With the increase in the concentration of IL-10, and gradually discreasing the volume of IL-2mRNA expression(P<0.05).The maximal concentration of IL-10 was 10ng/ml.CONCLUSION:①IL-10 inhibits IL-2,TNF-αproduced by PBMC and this effect was dependent on the concentration of IL-10. The maximal concentration of IL-10 was 10ng/ml.②IL-10 as an immune measurement is hopeful to be a new breakthrough in the treatment for epilepsy.The mechanism of IL-10 is to inhibit the expression of cytokine mRNA, thereby reducing nerve cell sexual excitement.
