Objective To investigate the expression level of angiotensin II receptor gene in obesity mice in different ages and to study the function of Losartan in left ventricular hypertrophy and expression level of Angiotensin II receptor gene. Methods To selected 30 mice in one month age and 30 mice in two month age,they were randomized to two groups:normal control group and obesity group,these mice were given high fat feed and common roughage respectively.After obesity model was built ,the obesity was divided into two groups:Losartan group,non-mediation group.Losartan group was given Losartan 10mg/kg.d and non-mediation group was given the saline.The expression level of angiotensin II receptor gene was detected by reserve transcription polymerase chain reaction (RT-PCR) methods.The changes of histology and pathology were observed by pathological section.Results (1) In one month age AT1 receptor gene expression is higher in non-mediation group than normal control group and Losartan group;LVMI increased in non-mediation group and improved in Losartan group. (2) In two month age The results is same as one month age mice. Conclusion There is left ventricular hypertrophy in different ages obesity mice;AT1 recepter gene express abnormal in different ages obesity mice;AT1 receptor antagon can reverse left ventricular hypertrophy and inhibit the synthesis of AT1 receptor mRNA.
