Abstract/Summary:
OBJECTIVE To study various activation on the in-vitrouproliferation and cytotoxicity of CIK cells and provide advanced evidence of the immunotherapy to leukemia after chemistrytherapy.MATERIAL AND METHODS Separate MNC from bone marrow or peripheral blood of 15 patients with ALL, 8 patients with AML and 3 patients with CML after CR by chemistry therapy. They were divided into three groups and cultured with different ways (CIK, PHA-CIK and PHA-Ag-CIK group). The cytokines were supplemented every three days. All the cells were harvested at the fourteenth days. The morphology of CIK cells were detected by using microscope and invert microscope. The phenotypes of CIK cells were analyzed by using flow cytometry and the killing tumor cell activity of CIK cells were tested with MTT method. The level of the IL-2, IFN-γ, which the harvested cells secreted were analyzed with ELISA.RESULT All of the three groups can were successfully induced to CIK cells. The proliferation rate and cytotoxicity of the PHA-CIK and PHA-Ag-CIK were stronger than the CIK cells.CONCLUSION PHA-CIK cells group had stronger proliferation and cytotoxicity than CIK cells group. The result provides an experiment basis for Anti-leukemia biotherapy.
