Abstract/Summary:
Objective: To investigate the apoptotic effect of the breast carcinoma Bcap-37 and the the expression of related gene p53, bax, bcl-2 induced by the extractive of Uighur medicine galla (MSZ), to determine whether MSZ has anticarcinoma effect in vitro and its mechanism. Methods; 1. To observe the Morphologic changes of Bcap-37 cells cultured in vitro with different concentration of MSZ after 48 hours under inverted microscope; to mensurate the inhibitive effect of Bcap-37 celis induced by MSZ through MTT method. 2. Apoptotic rate and cell cycles of Bcap-37 cells cultured in vitro with different concentration of MSZ after 48 hours were detected by flow cytometer. 3. The level of related Gene p53 mRNA, bax mRNA, bcl-2 mRNA was evaluated by RT-PCR methods. Results: 1. Under inverted microscope after 24 hours, it can be observed that a number of apoptotic cells had representative morphological characteristics to be round, large, or small, the cell membrane crimpled, and separated with neighboring cells, after 48hours the change of the Bcap-37 cells were of much more distinct. When dyed with HE, Bcap-37 cells appears to be round or anomaly, the bulk increased or reduced, some cells' membrane became unintegrity, even some cells' nucleolus disintegrated; tested by MTT, the inhibitive rate of Bcap-37 cells go up with the concentration increase of MSZ. 2. Apoptotic peak appeared in histogram of the flow cytometer in cells of test groups, while apoptotic peak did not show in cells of the blank group, and the differences of apoptotic rate between test groups and the negative group were significant (p<0.05).
