Objective: Ulcerative colitis(UC)was a precancer lesion of colorectal carcinoma, but the pathological mechanism was not clear. Most domestic reported cases which were diagnosed lacked of specific histopathological feature. So it was easily misdiagnosed. The purpose of our study is to detect the immune characteristics , change of mucinous secretion and histopathological features by immunohistochemistry(IHC) stain, mucinous histochemistry stain and HE stain,in order to get a better understanding on UC and provide some useful diagnostic indexes. Methods: UC and controls were studied to know the immune characteristics, change of mucinous secretion and histopathologic features. 1. Immune characteristics: The expression of CD45RO,CD20,CD68 were detected by IHC stain to know the changes of T, B lymphocytes and macrophages. The change of eosinophil infiltration was analyzed in HE stain slides; 2. Mucinous secretion:MUC2 was examined by IHC stain, The changes of sulphation and sialyl of mucus were examined by mucinous histochemical stain (HID-AB); 3. Histopathological feature of dysplasia was analysed in HE stain slides. Results: l:Immune characteristics: The positive rates of CD45RO and eosinophil infiltration showed significant difference among three stages (P<0.01) , they were higher in active stage than in controls(P<0.01) ; The positive rates of CD20 and CD68 showed significant difference among three stages(P< 0.01) . They were higher in active stage , inactive stage than in controls. 2.The change of mucinous secretion: The positive rate of MUC2 did not show significant difference among three stages (P>0.05).
