The gingko leaf is a kind of latent resources, There are many kinds of functional components mainly includes the flavones, the alkaloid, the multi- carbohydrates, the phenol, the amino acid, the trace element and so on, it has the broad development prospect. Our country gingko leaf output accounts for the world output about 70%, Studies on isolation, purification and hypoglycemic activity of polysaccharides from gingko leaves are in this article.We determined gingko leaf polysaccharide content by the phenol sulfuric acid method and the anthrone sulfuric acid method, finally indicated that absorbency Y and the polysaccharide content X of the anthrone sulfuric acid method between regression equation is Y=0.0076X+0.0071, correlation coefficient R~2=0.9995, the returning ratio is 100.26%, the accuracy is 0.069, the relative error is 0.51%. The result is better than the phenol sulfuric acid method, thus we think the anthrone sulfuric acid method determining gingko leaf polysaccharide content more ideal method, When we do purification experiment of gingko leaf polysaccharides, we may use it for the partial collections of polysaccharide and carry on the track examination.The best extracting technology of the gingko leaf polysaccharide was decided by the single factor and the orthogonal experiments :grinding degree is 60 mesh, the temperature is 100℃, the time is 4 hours, raw material and water ratio is 1:30.Sevag method, trichloroacetic acid method, Isoelectric point method was the compared by experiments. The best deproteinating method and condition was obstained. the sevag condition is that the chloroform and the normal butyl alcohol proportion are 4: 1, the sample fluid and the sevag volume compares 3: 1. We added sevag liquild into the sample fluid according to the condition .Polysaccharides was purified by the ionic exchange column DEAE cellulose and the Sephacryl-200, we obstrained the polysaccharide product. We analyzed the product with infrared ,the highly performance liquid chromatography and the gas chromatography , results in that the polysaccharide purity above 94%, the molecular weight is 12,749. includes: the rhamnose, the arabinose, the mannitose, the glucose, the galactose.
