OBJECTIVE To investigate the effect of artemether on the expression of IL-10 and IL-12 in HaCaT cells after exposure to UV ,and to observe the photo-protective effect of artemether on HaCat cells after UV irradiation.METHODS Subconfluent HaCat cells were shammed or irradiated with UVAof 2.00J/cm~2 or UVB of 60.00mJ/cm ~2 respectively. The cells were subsequently treated with hydroxychloroquine and different concentrations of artemether. At 12h after irradiation , the expression of IL-10R and IL-12R in HaCat cells were examined by immunohistochemistry(S-P method) and the percentage of positive cells were calculated. The secretion level of IL-10 and IL-12 were measured by ELISA. RESULTS Similar to hydroxychloroquine, artemether has protective effect on HaCaT cells damaged from UV irradiation. Both UVA and UVB can increase the expression of IL-10R, IL-12and IL-12R in HaCaT cells. Different wavelength of ultraviolet has different effect on the secretion level of IL-10. Different effect of hydroxychloroquine and artemether on the expression level of IL-10 and IL-12 were observed. The effect of artemether is dose-dependent.CONCLUSIONS Artemether has photo-protective effect on HaCaT cells cultured in vitro probably through regulating the level of IL-10 and IL-12 .
