Effect Of Litsea Cubeba Root Decoction On The Serum Levels Of TNF-α, IL-1β And The Synovitis In Rats With Collagen Ⅱ-induced Arthritis(CIA)

Objictive To investigate the effect of Litsea cubeba root decoction on the serum levels of TNF-α, IL-1β and the synovitis in rats with collagen II-induced arthritis(CIA) .To examine the therapeutice effect of Litsea cubeba root decoction on the collagen II-induced arthritis(CIA) in rats and explore its related immunological mechanisms.Methods The rat experimented arthritis model was established by subcutaneous injection with collagen II from chicken; Rats were distributed into seven groups randomly: normal control group , model control group , Tripterygium glycosides group, prednisone group,low-,middle- and high-dosage Litsea cubeba groups; The severity of arthritis was evaluated by the Arthritis Index; The serum TNF-α and IL-1β levels were detected with ELISA; HE pathological sections of synovial tissue were prepared to observe the influences of Litsea cubeba root on inflammatory infiltration, hyperplasia of type A macrophage(MA), hyperplasia of fibrous tissue(FT) and hyperplasia of synovial cell in model rats with collagen induced arthritis.Results The incidence of arthritis in the rats immunized with collagen II was approximately 68.2% (male rats 52.7%, female rats 83.6%); After 24 days drug administering, the Arthritis Index of middle- and high-dosage Litsea cubeba groups were notably lower than those of model control group (p<0.01)and no significantly difference with prednisone group and Tripterygium glycosides group; The serum TNF-α and IL-1β levels of the model group , Tripterygium glycosides group, prednisone group,Litsea cubeba root groups were notably higher than those of the normal group (p<0.001); The serum TNF-α and IL-1β levels of the Tripterygium glycosides group, prednisone group, Litsea cubeba root groups were notably lower than those of the model control group (p<0.001) ,and there was a probably dose-dependent relationship between the increasing of dosage of Litsea cubeba root and the decreasing of IL-1β levels;...
