Objective: Aim of present study was to explore the preventive effect of promethazine on reperfusion injury after cerebral ischemia and the course of apoptosis of neuron, and provide some evidences to find new therapeutical method.Material and Methods: (1)Following Longa' s method with modification, SD rats were selected to make reversible MCAO models, rats were randomly divided into sham-operated group, 5 control groups(reperfusion for 1h, 2h,6h,1d,3d,7d following MCAO for 2h) and 5 Promethazine groups with the same time point as control groups. Neurologic outcome was scored on a six-point scale. the infarction volume were measured by TTC staining .Continually to observe the anatomyic and functional changes of rats' brain after cerebral ischemia. (2)Rats with brain ischemia and reperfusion models were made by reopening both side common carotid artery after clipping it 10 minutes and were assigned to 3 group: sham-operated treated with Promethazine(S+P), reperfusion after ischemia treated with Promethazine(IR+P),and reperfusion after ischemia treated with 0.9%NaCl, the first and second groups were divided again 3 groups according the different concentration of Promethazine:A. 5mg/kg B. 15mg/kg C. 45mg/kg. we compared MPT in different groups. Results: (1) In control groups, the rats had the most severe disorders in neurological function after reperfusion for 1—3d following MCAO 2h, and began to recover after reperfusion for 7d.
