The aims of this study were to investigate the quality specification of morphine-carrier protein conjugate, and observe the pharmacological actions of murine anti-morphine monoclonal antibody (McAb) in vivo. The work probably could prepare for the development of human morphine vaccine and fully human anti-morphine McAb.1. Characterization of morphine-carrier protein conjugates:The research examined the purity of conjugate by SDS-PAGE, and determined the amount of protein, and the molecular ratio of protein to morphine by UV. The results demonstrate that the purity of conjugate, prepared by the prior synthetic route, is high; and yield is nearly invariable. The molecular ratio of bovine serum albumin (BSA) to morphine is 1:36, and blue carrier immunogenic protein (BC) to morphine is 1:271.2. Preparation of anti-morphine McAb and its pharmacological evaluation: Firstly, we prepared a large amount of anti-morphine McAb, and then weinvestigated its pharmacological actions.(1) Effects of anti-morphine McAb on morphine's analgesic action by hot plate test.
