Objective:The purpose of these experiments was to investigate the inhibition of the antioxidant—ginsenoside—Rg1on the rabbit lens epithelial cell apoptosis induced by H2O2 and to provide basic methods and experimental basis for the mechanism of medication treatment of the cataract。Methods:Rabbits lens in vitro were placed in 6—well flat bottomed tissue culture plates and incubated in the Dulbecco's modified eagle's nedium (DMEM)for 72 hours and were divided into four groups;control group(exclusive of H2O2),H2O2 group(involving0.3Mm ),H2O2 added Rg1group and H2O2 added taurine group。The transparency of all lenses was detected after above lenes which were incubated for 3h,6h,12h,18h,24h,36h,48h and 72h;at the same time,the histological changes of all lenses which were incubated for 72 houes were examined by electron microsopies and verified the occurrence of programmed cell death and the expression of bcl--2 and bax was detected by the immunohistochemical streptavidin—peroxi—dase ( SP ) method , as well as by terminal deoxyribonucleotide transferase—mediate dUTP nick end labeling(TUNEL)detection,we examined the affect of Rg1 on the number of H202—induced rabbit lens epithelial cell with apoptosis ,the related comparisons and statistic analyses were carried out。Results:1,The longer time incubated in the DMEM/F12 exclusive...
