Objective:1,To study the effect of three different inhibitors of glycosphingolipid biosynthesis D-PDMP,D-PPMP and NB-DNJ on SHG cells growth.2,To inverstigate D-PDMP exerted the inhibitory effect on cell growth through inducing apoptosis.3,To inverstigate the possible mechanism of the inhibitory effect of D-PDMP on SHG cells growth.Method:1,The effect of three different inhibitors of glycosphingolipid biosynthesis on SHG cells growth was measured by MTT assay.2,Apoptosis was measured by AO/EB staining, TUNEL and flow cytometry.3,The concentration and component of ganglioside were measured by HPTLC.4,The concentration of ceramide were measured by DAGK assay.5,The concentration of sphingomyelin were measured by HPLC.Result:1,D-PDMP exerted a dose-dependent inhibitory effects on SHG cells growth , D-PPMP NB-DNJ did not affect cellular biosynthesis.2,AO/EB staining and TUNEL showed characterisitic apoptosis morphologicalla performed, and the result of flow cytometry showed that a xub-G1 peak appeared before the normal diploid peak after treating the SHG cells with high concentration D-PDMP.
