Objective To investigate the proliferation and differentiation of bone marrow stromal cells (BMSCs) from the normal rats and the osteoporosis rats, and to evaluate the effects of 17β-estradiol (E2) and raloxifene(Ralo) on the proliferation and differentiation of the two groups. Methods 1.Thirty-two three-month-old female SD rats were randomly divided into four groups: ovariectomized rats( the OVX ), sham-ovariectomized rats( the Sham ), the OVX rats treated with E2( the OVX + E2 ), and the OVX rats treated with Ralo( the OVX + Ralo ). One week after the ovariectomy, the rats were treated with the medicine. 2.Eight weeks after the operation, we investigated the differences between the four groups by examining the body weight, the bone mineral density(BMD), histological examination and serum levels of biochemical indices 3.At the same time, under the sterile condition, we collected the thighbone and the tibial bone, and then gained the BMSCs, cultured them in vitro. 4. We used two concentration E2 or Ralo to treat the BMSCs from the Sham and the OVX. Cells between the 3rd and 4th passages in culture have been used for this study. We evaluated the proliferation of the cells by MTT and the differentiation by comparing the synthesized alkaline phosphatase(ALP). Results 1. The BMD of total body of the OVX were significantly lower than the other three groups(P<0.01), and the BGP concentration of the OVX were obviously higher than the others(P<0.01 or P<0.05). And there was no statistical difference between the BGP concentration of the OVX+E2 ,the OVX+Ralo and the Sham's(P>0.05).
