Object: To investigate the prevention of L-arginine and Losartan respectively and their associated on intimal hyperplasia of autografted venous of rats, then discuss their potential mechanism and possible synergistic action.Methods: The establishment of autografted venous model was used with external jugular vein- arteria carotis transplantation in rats. According to the administration condition after autografted venous transplantation, 80 SD rats were randomly classified into four groups: the control group(A) L-arginine-treated group(B)Losartan-treated group(C) and combination treated . Before feeding to rats and at 0, 2, 4, 6, 8 week after transplantation , NO content in blood serum were detected . At 0,2,4,6,8 week after transplantation, the vein grafts were harvested to carry out histopathologic examination and the expression of PNCA , cyclinA , VCAM-1, eNOS by immunohistochemistry. Observation and analysis of ultramicrostructure change of transplantation vein by electron microscope.Results: Both L-arginine and Losartan degrade auto grafted venious intimal hyperplasia, restrain vascular smooth muscle cell migrate to intima and vascular smooth muscle cell proliferation, lessen extracellular matrix apposition and Inflammatory cell infiltration. L-arginine is more effective than Losartan in tunica media and adventitia of auto grafted vein. In combination treated group, the degree of auto grafted venious intimal hyperplasia, vascular smooth muscle cell migrate to intima and vascular smooth muscle cell proliferation, extracellular matrix apposition and Inflammatory cell infiltration are minimum. L-arginine and Losartan improve NO content in blood serum. In L-arginine-treated group, the level that NO content in rats blood serum inprove is higher than in Losartan-treated group.
