As an innovative treatment for a range of cancers, photodynamic therapy (PDT) has received extensive attention. The over-all efficacy of the treatment depends greatly on the behavior of the photosensitizer, including its selectivity toward cancer cells and efficiency to generate reactive oxygen species(such as singlet oxygen). Owing to the high efficiency to generate reactive oxygen species, phthalocyanines(Pcs) have been attended increasingly as photosensitizers. However, the selectivity of phthalocyaniens for cancer cells isn't perfect, so how the selectivity can be further enhanced is one of focus in the filed of PDT. Recently, It has been found that Pcs which conjugate with receptor-mediated proteins can improve effectively targeting to some cancer cells. In this dissertation, a series of silicon(Ⅳ) phthalocyanines axially modified by alkoxy groups have been synthesized; and conjugations of Pcs with protein have also been investigated. It is hoping to offer some interesting information on designing new targeting photosensitizer compounds. The main results of this paper are summarized as following.1) Eleven silicon phthalocyanines axially substituted have been synthesized, including a dicationic one. The axial substituted groups contain one and more ethoxyl, two of these contain N-heterocyclic as well. All compouds were characterized with HPLC, IR, UV, MS, NMR together with elemental analysis.2) Some physical and chemical properties involved PDT have been investigated, including solubility, stability and molecule spectrum.The results show that the increase of axial glycol can improve the hydrophility of phthalocyaninatosilicon; Bis(2-(N-methyl-morpholine) ethoxy) phthalocyaninatosilicon diiodide (ZSDI), axially substituted by cation, not only can dissolve in water, but exist in the form of monomer with theλmax of 683nm which is helpful for photodynamic therapy.3) Interaction of nineteen Pcs with three protein (BSA,HSA,apoTf) have been studied by fluorescence spectral analysis. The binding constants have been calculated by Stern-Volmer equation and static quenching methods, and the effect of Pc's structure has been discussed.4) The 41 conjugates were also prepared by incubating a mixture of phthalocyanines and the corresponding serum albumin followed by separation by gel permeation chromatography.
